Fester's Quest - Boss Glitch
Have a key in your inventory and selected, move across the room going down+left, and mash the B button. Thing gives you invis potions, boss dies, screen glitches, and you're sent to the next area. -

@Jaayyy1993: You are pressing the gravity switch too late. You've got to press it just as the left slab touches the top of the screen. That way the left one will stick to the ceiling and the right one will not.

It's pretty risky (or at least, I fail half the time i try it). I think it saves about 12 seconds. For a casual speedrun, i wouldn't recommend it. For a really serious speedrun, 12 seconds is worth it. In my opinion, at least.
Limbo trick - Jumping the tire
Rather than backtracking to wait for the tire to roll into the water, you can actually jump it. It's possible to jump it facing right, but I find that it's easier to do a stationary jump facing le... -
